R-spondins constitute a family of secreted factors (R-spondin-1, R-spondin-2, R-spondin-3, R-spondin-4), all of which have been reported to act as general regulators of canonical Wnt signaling. R-spondins require Wnt ligands and LRP6 for activity and amplify signaling of Wnt-3a, Wnt-1, and Wnt-7a. The factors also antagonize dkk-1 activity by interfering with dkk-1 mediated LRP6 and Kremen association (Kim et al, 2008).
