Datta et al (2007) have reported that HTLV-I p30 protein interferes signaling through the Toll-like receptor TLR4. p30 interacts with and inhibits the DNA binding and transcription activity of PU.1, which results in the downregulation of TLR4 expression from the cell surface. Expression of p30 hampers the release of pro-inflammatory cytokines MCP-1, TNF-alpha, and IL8 and stimulates release of anti-inflammatory IL10 following stimulation of TLR4 in human macrophages. p30 also increases phosphorylation and inactivation of GSK3-beta, a key step for IL10 production.
