A technique used to detect hematopoietic stem cells (see also: hematopoiesis).
This assay is a short-term suspension culture assay in which potentially colony-forming cells (see: Colony formation assay) are grown first in liquid culture for one week before they are replated in semi-solid medium.
Numbers of colonies obtained in this way are considered as an indicator of the number of hematopoietic progenitors of an early stage (earlier than those normally obtained after 14 days of growth in semi-solid medium).
Interpretation of the two-step liquid culture assay is based on the assumption that hematopoietic progenitor cells normally not detected in a colony formation assay
