This sequence motif, termed DRY motif by Graham (2009), is found in the second intracellular loop of signaling chemokine receptors. This motif, or subtle variants of it, is responsible for G-protein coupling to the receptors (Kraft et al, 2001; Damaj et al, 1996). The sequence motif is absent/altered in some atypical chemokine receptors. These receptors are characterised by promiscuity of ligand binding and an apparent inability to signal following ligand binding and, in some cases, by an ability to internalise and degrade chemokine ligands, and are thus thought to serve as chemokine reservoirs, scavengers, or decoy receptors.
