The acronym DN stands for "double-negative". This refers to a population of immature T-cells (thymocytes) that do not express the CD antigens CD4 or CD8. DN3 cells are a subpopulation of these cells (see also: DN1 cells) with the phenotype CD4(-), CD8(-), CD25(+), CD44(-). It is at this stage that commitment to T-cells occurs. These cells give rise to double-positive thymocytes that CD4 and CD8 that develop further into mature T-cells (Huesmann et al, 1991; Ceredig and Rolink, 2002).
