Cytomegalovirus US2 protein

This immediate-early gene product encoded in the genome of human cytomegalovirus suppresses antigen presentation to CD8(+) cytotoxic T-lymphocytes and therefore acts as an immunoevasin, hindering viral clearance by cytotoxic T-cells. US2 utilizes the cellular E3 ligase TRC8 [translocation in renal cancer from chromosome 8] to ubiquitinate and subsequently degrade MHC-1 (Stagg et al, 2009). US2 also targets the MHC-2 antigen presentation pathway, cooperating with Cytomegalovirus US3 protein (Hegde et al, 2002; Tomazin et al, 1999), initiating the retrotranslocation of MHC-2 DR-alpha chain and DM-alpha chain from the endoplasmic reticulum back to the cytosol for subsequent degradation.
