Flow cytometry offers a way to detect cytokines at the single cell level by means of fixation and permeabilization of cells with subsequent specific intracellular staining with the use of labelled monoclonal anti-cytokine antibodies. The technique also allows determination of cytokine production phenotypes by the simultaneous detection of two or more cytokines within a single cell. This is particularly useful, for example, for the identification of distinct T-helper cell subsets of lymphocytes (Th1 cells, Th2 cells) that differ in their cytokine production profiles.
