The crystal structures of several cytokines and hormones with diverse biological activities and very little sequence homology reveal a common overall topology that is not deducible from sequence comparisons because the homology among these proteins is generally low. Cystine knot structure are conserved among divergent species and most likely evolved in extracellular signaling molecules of multicellular organisms. Proteins with cystine knots have not been found in the unicellular yeast genome but are present in nematode, fly, and higher species.
