This homodimeric protein is a disintegrin isolated from the venom of Agkistrodon contortrix contortrix (Southern Copperhead), identified originally as an agent that inhibits platelet aggregation (Trikha et al, 1994). Each chain of the protein contains an RGD sequence motif that modulates its interaction with integrins on tumor cells and angiogenic vascular endothelial cells. The protein is a potent inhibitor of angiogenesis and tumor growth. A recombinant soluble monomeric form of Contortrostatin is also active in an orthotopic, xenograft model of human breast cancer (Minea et al, 2005).
