Claudin-8 is a member of a large multigene family members of which are known to interact with a number of proteins and are involved in signal transduction to and from the tight junction (Krause et al, 2008). Claudin-8 acts primarily as a cation barrier and plays an important role in the paracellular cation barrier of the distal renal tubule (Yu et al, 2003). Angelow et al (2006) have reported that the induction of claudin-8 expression reduces permeability to protons, ammonium, and bicarbonate. Claudin-8 probably functions to limit the passive leak of these three ions via paracellular routes, thereby playing a permissive role in urinary net acid excretion.
