This peptide, identified by Samoylova and Smith et al (1999) through phage display selection, is a homing peptide that shows enhanced in vivo skeletal and cardiac muscle binding and targets myofibers in mice. When used with an Adeno-associated virus (AAV) vector, systemic delivery of this vector modified with the muscle-targeting peptide improves muscle tropism (Yu et al, 2009).
Yin et al (2013) have used this sequence as a muscle-specific targeting sequence together with a cell-penetrating peptide sequence linked in a construct to morpholino phosphorodiamidate oligomers. This conjugate improves cellular uptake by muscle cells and has been shown to enhance the cellular delivery of oligonucleotides.
