This protein complex consists of mitochondrial adenylate kinase 2 (AK2), FADD, and caspase-10. Complex formation has been shown to constitute a cell death pathway of intrinsic apoptosis that may be involved in tumorigenesis (Lee et al, 2007).
Downregulation of the expression of AK2 attenuates cell death by apoptosis induced in human cells by treatment with etoposide or staurosporine. This does not affect cell death signalled through processes involving TRAIL or FAS ligand.
During intrinsic apoptosis, AK2 translocates to the cytoplasm, whereas this event is diminished in cells
